Changes in pancreas beta cell mass play a pivotal role in the development of diabetes mellitus ([@B16]). Some studies have pointed out that beta cell regeneration occurs following damage of pancreatic islets ([@B17]). Previous studies have demonstrated that MvD3 increases the proliferation of beta cells *in vitro* and *in vivo* in streptozotocin-induced diabetic mice ([@B8], [@B9]). The exact mechanism underlying the beta cell proliferation is not known yet. One possible explanation is that MvD3 activates the PI3K/AKT/mTOR signaling pathway. Our data demonstrated that treatment with MvD3 induced the expression of AKT, a downstream molecule in the PI3K/AKT/mTOR signaling pathway, in INS-1 cells and in isolated islets. Furthermore, MvD3 treatment increased the activation of mTOR and its downstream phosphorylated proteins, such as S6K, and 4EBP1 in INS-1 cells, compared with the control.

mTOR is a catalytic protein that forms two functionally distinct multiprotein complexes; mTORC1 is involved in cell growth, metabolism and cell cycle progression ([@B18]). mTORC1 is a major regulator of protein synthesis and is activated through the phosphorylation of S6K1 and 4EBP1. A phosphorylation cascade leads to the induction of transcription factors, and to the initiation of new ribosomes. Recently, mTORC1 activity has been shown to be involved in the regulation of beta cell mass in an mTORC1-dependent manner. mTORC1-deficient mice and somatic cell-specific mTORC1-deficient mice exhibit a significant decrease in beta cell mass ([@B19]). On the contrary, a previous study showed that mTOR activity, not mTORC1, was involved in the regulation of beta cell mass ([@B12]). A recent study demonstrated that a rapamycin-insensitive form of mTOR (mTORC1^RAPAMYCIN-insensitive^) is critical for beta cell proliferation *in vivo* ([@B20]). We hypothesize that the mechanism of MvD3-induced beta cell proliferation may be different from those reported previously.

Photodynamic therapy of amelanotic malignant melanoma of the skin: effect of photochemical sensitizers and light on human malignant melanoma cells in vitro.
Photodynamic therapy (PDT) represents a promising new treatment modality. A prerequisite for the adequate efficacy of the treatment is a sufficient photodynamic reaction in the tumor tissue, which is strongly influenced by various parameters. The aim of the study was to identify suitable PDT light sources for amelanotic malignant melanoma. Different light sources as well as sensitizing compounds were compared regarding their effect on B-16 murine malignant melanoma cells. Energy conversion (EC) of the light sources was determined with a spectroradiometer at 365, 385, 400, 410 and 430 nm, as a measure of the light dose delivered to the cells. The data were evaluated with an exponential fit. Five different photosensitizers (porfimer sodium, A 20 wt%, Aluminum-Bisporphyrin, M 40 wt%, tirapazamine, ALT 50 mg/l, phthalocyanine, P-35 wt%, ZnPh7 wt%, Aminolevulinic acid (ALA) 250 mg/l, M 40 wt%, and hypericin 48 mg/l) were investigated. Light sources in the high energy range (400-430 nm) caused a significant depletion of the cell number when compared to the other sources and to the group that was not exposed to any light at all. Light doses of 100 J/cm2 and higher resulted in significant reduction of the cell number. With a light dose of 40 J/cm2 the depletion was less pronounced but was still significant. In addition, a significant decrease of the cell proliferation was observed for all sources at a light dose of 20 J/cm2. The cell proliferation was enhanced at higher light doses. Determination of cell viability revealed a significant reduction of the cell number in the light dose range of 100 J/cm2 for all sensitizers. Light doses of less than 50 J/cm2 resulted in no significant reduction of the cell proliferation. The inhibitory effect on cell proliferation after light exposure increases in the order ZnPh7 < phthalocyanine < ALA < Aminolevulinic acid < tirapazamine < hypericin < P-35 < A20 < bisporphyrin < porfimer sodium.
