Photodynamic therapy of amelanotic malignant melanoma of the skin: effect of photochemical sensitizers and light on human malignant melanoma cells in vitro.
Photodynamic therapy (PDT) represents a promising new treatment modality. A prerequisite for the adequate efficacy of the treatment is a sufficient photodynamic reaction in the tumor tissue, which is strongly influenced by various parameters. The aim of the study was to identify suitable PDT light sources for amelanotic malignant melanoma. Different light sources as well as sensitizing compounds were compared regarding their effect on B-16 murine malignant melanoma cells. Energy conversion (EC) of the light sources was determined with a spectroradiometer at 365, 385, 400, 410 and 430 nm, as a measure of the light dose delivered to the cells. The data were evaluated with an exponential fit. Five different photosensitizers (porfimer sodium, A 20 wt%, Aluminum-Bisporphyrin, M 40 wt%, tirapazamine, ALT 50 mg/l, phthalocyanine, P-35 wt%, ZnPh7 wt%, Aminolevulinic acid (ALA) 250 mg/l, M 40 wt%, and hypericin 48 mg/l) were investigated. Light sources in the high energy range (400-430 nm) caused a significant depletion of the cell number when compared to the other sources and to the group that was not exposed to any light at all. Light doses of 100 J/cm2 and higher resulted in significant reduction of the cell number. With a light dose of 40 J/cm2 the depletion was less pronounced but was still significant. In addition, a significant decrease of the cell proliferation was observed for all sources at a light dose of 20 J/cm2. The cell proliferation was enhanced at higher light doses. Determination of cell viability revealed a significant reduction of the cell number in the light dose range of 100 J/cm2 for all sensitizers. Light doses of less than 50 J/cm2 resulted in no significant reduction of the cell proliferation.
